This can be done directly on the graph or with curve fitting software which is typically found on ELISA plate readers. If a quantitative result is needed, the simplest way to proceed is to average the triplicate of the standards readings and deduct the reading of the blank control sample.

Next, plot the standard curve, find the line of best fit or at least draw a point to point curve so that the concentration of the samples can be determined. Any dilutions made need to be adjusted for at this stage. This is generally the practical extent to which manual calculation can be taken. Using linear regression within a software package adds several more checking possibilities; it is possible to check the R2 value to determine overall goodness of fit.

Accuracy can then be further enhanced by using further standard concentrations in that range. One aspect of the linear plot is that it compresses the data points on the lower concentrations of the standard curve, hence making that the most accurate range area most likely to achieve the required R2 value. To counteract this compression a semi-log chart can be used; here the log of the concentration value on x-axis is plotted against the readout on y-axis.

This method gives an S-shaped data curve that distributes more of the data points into the more user friendly sigmoidal pattern. For example, the production of viral vaccinesrecombinant proteins using viral vectors and viral antigens all require virus quantification to continually adapt and monitor the process in order to optimize production yields and respond to ever changing demands and applications.

Examples of specific instances where known viruses need to be quantified include clone screening, multiplicity of infection MOI optimization and adaptation of methods to cell culture. This page discusses various techniques currently used to quantify viruses in liquid samples.

These methods are separated into two categories, traditional vs. Traditional methods are industry-standard methods that have been used for decades but are generally slow and labor-intensive. Modern methods are relatively new commercially available products and kits that greatly reduce quantification time. This is not meant to be an exhaustive review of all potential methods, but rather a representative cross-section of traditional methods and new, commercially available methods.

While other published methods may exist for virus quantification, non-commercial methods are not discussed here. Plaque-based assays are the standard method used to determine virus concentration in terms of infectious dose.

Virus quantification

Viral plaque assays determine the number of plaque forming units pfu in a virus sample, which is one measure of virus quantity. This assay is based on a microbiological method conducted in petri dishes or multi-well plates. Specifically, a confluent monolayer of host cells is infected with the virus at varying dilutions and covered with a semi-solid medium, such as agar or carboxymethyl celluloseto prevent the virus infection from spreading indiscriminately.

A viral plaque is formed when a virus infects a cell within the fixed cell monolayer. The infected cell area will create a plaque an area of infection surrounded by uninfected cells which can be seen with an optical microscope or visually pouring off the overlay medium and adding a crystal violet solution for 15 minutes until it has colored the cytoplasm, gently removing the excess with water will show uncolored the location of dead cells [2].

Plaque formation can take 3—14 days, depending on the virus being analyzed. The focus forming assay FFA is a variation of the plaque assay, but instead of relying on cell lysis in order to detect plaque formation, the FFA employs immunostaining techniques using fluorescently labeled antibodies specific for a viral antigen to detect infected host cells and infectious virus particles before an actual plaque is formed.

The FFA is particularly useful for quantifying classes of viruses that do not lyse the cell membranes, as these viruses would not be amenable to the plaque assay. Like the plaque assay, host cell monolayers are infected with various dilutions of the virus sample and allowed to incubate for a relatively brief incubation period e.

Plates are subsequently probed with fluorescently labeled antibodies against a viral antigen, and fluorescence microscopy is used to count and quantify the number of foci. The FFA method typically yields results in less time than plaque or fifty-percent-tissue-culture-infective-dose TCID 50 assays, but it can be more expensive in terms of required reagents and equipment.

Assay completion time is also dependent on the size of area that the user is counting. A larger area will require more time but can provide a more accurate representation of the sample. Fifty-percent tissue culture infective dose TCID 50 is the measure of infectious virus titer.

This assay may be more common in clinical research applications where the lethal dose of virus must be determined or if the virus does not form plaques. When used in the context of tissue culture, host cells are plated and serial dilutions of the virus are added. After incubation, the percentage of cell death i. This method can take up to a week due to cell infectivity time. But the following reference defines the relationship differently: Assuming that the same cell system is used, that the virus forms plaques on those cells, and that no procedures are added which would inhibit plaque formation, 1 ml of virus stock would be expected to have about half of the number of plaque forming units PFUs as TCID In some instances, two or more plaques might by chance form, and thus the actual number of PFUs should be determined experimentally.

Mathematically, the expected PFUs would be somewhat greater than one-half the TCID 50since the negative tubes in the TCID 50 represent zero plaque forming units and the positive tubes each represent one or more plaque forming units. A more precise estimate is obtained by applying the Poisson distribution. Therefore, one could multiply the TCID 50 titer per ml by 0.

No problem. This is where things can get interesting. In order to determine a quantity of something you will need to compare your sample results to those of a set of standards of known quantities. The standards, in your assay, should be tested at a range of concentrations that yields results from essentially undetectable to maximum signal. You can think of the standard curve as the ideal data for your assay.

Once the standard curve is generated it is relatively easy to see where on the curve your sample lies and interpolate a value. The goal is to determine values of m and c which minimize the differences residuals between the observed values i. To check the predicted fit of the line one usually calculates all the residuals observed — predicted and sums all the differences. The smaller the sum the better the data fit the predicted curve. However, since some observed values will likely be above the fitted curve and some below you will get positive and negative residuals.

Summing these is not very useful, as even a random set of data points may generate residuals that sum close to zero. To get around this you should square each of the residuals, which render all the values positive, then sum them.

This is known as the sum of squares SSq. The smaller the SSq, the closer the observed values are to the predicted, the better the model predicts your data. The good news is that linear regression is pretty easy. The bad news is that linear regression is seldom a good model for biological systems.

Four Parameter Logistic 4PL Regression This leads us to another model of higher complexity that is more suitable for many biologic systems. This model is known as the 4 parameter logistic regression 4PL. The model fits data that makes a sort of S shaped curve. The curve can only be used to calculate concentrations for signals within a and d. Samples outside the range of the determined a and d cannot be calculated. This model is a little trickier than the linear regression model above. So in addition to studying vocabulary words, you should also practice doing advanced reading and test your ability to define tough words based on their context. They've posted a number of free new SAT practice tests. Start there to get a sense of what the new SAT Reading section is like. Still have old SAT prep books sitting around. You can use old SAT Critical Reading questions to practice, but focus on the passage-based questions and ignore the sentence-completion questions. ACT Reading section questions will also be helpful, as they are all passage-based and contain vocabulary in context as well as logical progression questions.

Another unlikely but helpful source is ACT Science questions. ACT Science also has you break down charts, graphs, and evidence. Want a bit more structure for vocabulary in context. One of my favorite tools for learning vocabulary in context is a browser app called ProfessorWord.

This article alone has about a dozen SAT vocabulary words, according to ProfessorWord. The writing section is quite different on the new SAT. There is more emphasis on logic and expression of ideas, higher-level writing skills, and punctuation. This means that there are fewer grammar rules tested in isolation, which in turn means fewer "gotcha" questions on the new SAT Writing section. However, being aware of writing style, construction, and organization is more important, since you will now be working with longer passages.

Start your studying by learning English grammar rules by heart. Then give the SAT's official practice tests a try. In terms of additional practice questions, we recommend you use ACT English practice questions, as these are all passage-based, like the new SAT Writing questions are. You can also use old SAT Writing multiple-choice questions to test your grammar rule knowledge, but remember to be ready for passages. Finally, the more you read and write, the better you will get at spotting writing organization and style naturally.
